| Comments | Vector: pGEM-Teasy, 3 week-old plants (cultivar Hawaii 7996) were inoculated with water (control) or Ralstonia solanacearum (an incompatible strain) (Rs-inoculated). Roots and collars of control and Rs-inoculated plants were harvested 24 and 48 hours post inoculation. Poly(A) RNA specimen prepared from control plant samples were mixed at 1:1 ratio, and that prepared from Rs-inoculated samples were also mixed at 1:1 ratio. Tomato cDNA library was then constructed by subtracting the pooled Rs-inoculated specimen from pooled control specimen using CLONTECH PCR-Select cDNA Subtraction kit. |
