The composition of the mature coffee beans determines the final quality and processing properties of the coffee produced from them. Analysis of the differences in gene expression during coffee bean maturation may explain the genetic and environmental variations underlying coffee quality.
The transcriptome of coffee beans was analyzed at three developmental stages, namely immature (green), intermediate (yellow), and mature (red) stages. Sequencing transcripts collected over 120 million 150bp paired reads from three samples at each developmental stage. More transcripts were expressed at the yellow stage. As the beans matured, the type of highly expressed transcripts changed from those associated primarily with galactomannan, triacylglycerol (TAG), TAG lipase, 11S, and 7S-like storage proteins, and Fasciclin-like arabinogalactan protein 17 (FLA17) in mung beans to those associated with FLA1 in yellow beans and TAG storage lipase SDP1 and SDP1 in red beans -like transcripts in the red bean. This study provides a genomic resource that can be used to study the effects of environment and genotype on the transcriptome of coffee beans and to develop coffee varieties and production systems that are better adapted to provide quality coffee under climate change.
